202 The importance of LRRC8 ion channel in keratinocyte differentiation

چکیده

Epidermal regeneration relies on a balance between keratinocyte proliferation and differentiation. Both processes are accompanied by cell volume changes. The anion channel LRRC8 mediates regulation upon hypotonic stress in keratinocytes its essential subunit LRRC8A is mainly expressed the basal, transient amplifying population of human epidermis. In vitro differentiation experiments primary confirmed these findings, as upregulated during onset leading to bell-shaped expression pattern. Therefore, we asked whether also involved process changes keratinocytes. order understand role performed different loss-of-function strategies. First, used CRISPR/Cas9 edit HaCaT cells immortalized line NHEK/SV-TERT. contrast cells, NHEK-SV/TERT can differentiate after Ca2+ switch 2D show stratification epidermal equivalents. both lines, found aberrant 3D experiments. Next, siRNA reduce levels keratinocytes, which better simulate vivo This significantly reduced RNA protein ion activity. Surprisingly, knockdown did not interfere with As has short window high initiation, specifically targeted siRNA. Strikingly, indeed leads capability. Our work identifies an molecule differentiation, main function where needed for from

The Importance of the Dissociation Rate in Ion Channel Blocking

Understanding the relationships between the rates and dynamics of current wave forms under voltage clamp conditions is essential for understanding phenomena such as state-dependence and use-dependence, which are fundamental for the action of drugs used as anti-epileptics, anti-arrhythmics, and anesthetics. In the present study, we mathematically analyze models of blocking mechanisms. In previou...

The role of nkx2.5 in keratinocyte differentiation.

BACKGROUND Nkx2.5 is a homeodomain-containing nuclear transcription protein that has been associated with acute T-lymphoblastic leukemia. In addition, Nkx2.5 has an essential role in cardiomyogenesis. However, the expression of Nkx2.5 in the skin has not been investigated. OBJECTIVE In an attempt to screen the differentially regulated genes involved in keratinocyte differentiation, using a cD...

The keratinocyte growth-differentiation switch.

Growth/differentiation control of normal epithelial cells has been relatively understudied, because of the complexities involved in their cultivation and characterization. The present review is focused on progress in this area using the mouse primary keratinocyte system. This system reproduces under well defined culture conditions the switch between epithelial cell growth and differentiation wh...
